    MGMT Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody specific for an epitope mapping between amino acids 9-24 at the N-terminus of human MGMT, demonstrating reactivity with MGMT from mouse, rat, and human origins by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MGMT, also known as O6-methylguanine-DNA methyltransferase or O-6-alkylguanine DNA alkyltransferase (AGT), is a vital DNA repair enzyme that removes mutagenic and cytotoxic alkyl groups from the O6 position of guanine, preventing mutations caused by alkylating agents. MGMT interacts with the tumor suppressor protein p53, which regulates MGMT expression in response to DNA damage. Functional p53 upregulates MGMT transcription, enhancing DNA repair capacity, whereas mutant p53 can result in reduced MGMT levels and increased genomic instability. Additionally, MGMT activity and stability are influenced by interactions with proteins involved in post-translational modification, such as ubiquitin ligases that tag MGMT for proteasomal degradation, thus regulating cellular concentration. The balance between MGMT synthesis and degradation is critical for determining cellular sensitivity to DNA-damaging agents, impacting the effectiveness of certain chemotherapeutic drugs. Furthermore, MGMT may interact with DNA repair complexes to facilitate efficient repair of alkylated DNA lesions, underscoring MGMT′s central role in maintaining genomic integrity.
