AChRα7 Antibody (319) is a rat monoclonal IgG1 antibody that detects AChRα7 protein of mouse, rat, human, and avian origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and flow cytometry (FCM). AChRα7 Antibody (319) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. AChRα7, also known as NACHRA7, CHRNA7-2, or CHRNA7, is a critical component of the nicotinic acetylcholine receptor family, which plays a vital role in fast synaptic transmission. AChRα7 is a 502 amino acid multi-pass membrane protein that forms a homopentamer, allowing creation of a transmembrane aqueous pore essential for ion flow. AChRα7 is predominantly located at the postsynaptic membrane of the neuromuscular junction, where binding acetylcholine facilitates ion influx leading to muscle contraction. Proper functioning of AChRα7 is crucial for neuromuscular signaling, and dysregulation has been implicated in various neurological disorders. AChRα7 interacts with RIC-3, a nicotinic acetylcholine receptor-associated protein, which is important for proper folding and trafficking of the receptor, further underscoring AChRα7′s significance in maintaining synaptic integrity and function.

Hi! We have produced mouse cells where the last exons of the AChRα7 gene have been deleted (exons 7 to 10). Still a small portion (N-terminal) of the AChRα7 is synthesized and express on the cells. Will this antibody bind to that protein fragment?

Thank you for your question. AChRα7 Antibody (319): sc-58607 was raised against bacterially expressed large cytoplasmic domain of AChRα7 of chicken origin. The epitope region of the antibody is between amino acids 319-469 of accession number P22770. The exact binding region of this antibody within the antigen has not been mapped. What is the approximate amino acid region that this specific antibody is supposed to bind?

Thank you for your question. This antibody was raised against bacterially expressed large cytoplasmic domain of AChRα7 of chicken origin. We have not characterized the exact epitope of this monoclonal antibody.

We recommend using a directly conjugated primary antibody. This product, sc-58607, is available conjugated to FITC, AF488, and AF647.

Rated 1 out of 5 by from ProblemasGreetings, Yes, I would like to write a review on this new Rat Anti-Alpha 7 Nicotinic Acetylcholine receptor antibody. My experience with this antibody has been negative. I also have a work colleague, which has the discontinued polyclonal Alpha 7 Nicotinic Acetylcholine receptor antibody, and it is better than the new rat monoclonal Santa Cruz offers and promotes. Nevertheless, this does not mean that the polyclonal antibody was great either, she had to do Immunoprecipitation (IP) of the protein (using a total of 500 ug in her cell lysate) in order to obtain high enough concentration of the protein in macrophage and microglial lysates, to observe a band in her Western Blot. Her last IP using the new antibody did not even detect a band. She is using the Pierce ECL system, the Femto chemiluminescence system to detect HRP-conjugated secondaries. Both antibodies were used at a concentration of 1:500 in TBS-T and 5% powdered fat-free Milk, overnigt incubation at 4C. I have tried the new Rat-monoclonal Alpha 7 antibody in whole mouse brain lysate and cerebellum, using enormous quantities of protein (80 and 100ug total protein per well) in a normal Western Blot (not performing IP) and the signal is still extremely faint. I also looked at the protein left in the 'pellet' of the lysate, and the pellet showed also faint almost invisible amounts. The dilution I used was 1:200 in TBS-T and 5% powdered fat-free Milk, overnight incubation at 4C. I use the LI-COR Infrared system of detection, using secondary antibodies that are excited at IR wavelengths. I definitely feel that this is not a practical antibody for Western blotting. Its titer is extremely low and normally one would expect to add a max amount of 20-40ug of a cell lysate to observe proteins. The fact that my colleague did not obtain a signal performing an IP or that I could only detect a faint, non-quantifiable band by densitometry, makes this antibody really useless to us. My recommendation: not god for WB, increase the titer of this antibody at least 5X. Thank you for your attention, -Yancy

Rated 4 out of 5 by from ImmunofluorescenceI used these antibodies for IF in transfected with CHRNA7 cells. Gave a strong signal. The antibody worked in WB too, the major product was around 55KDa, but gave few unspecific smaller bands.
