IRE1α Antibody (B-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects IRE1 alpha protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). IRE1α (B-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IRE1 alpha protein plays a crucial role in the unfolded protein response (UPR), a cellular stress response activated when misfolded or unfolded proteins accumulate in the endoplasmic reticulum (ER). This response is vital for maintaining cellular homeostasis and preventing apoptosis under stress conditions. IRE1α, along with IRE1β homolog, is localized in the ER lumen and features a transmembrane domain spanning the ER membrane, as well as a cytosolic Ser/Thr kinase domain. Upon activation, IRE1α undergoes oligomerization and trans-phosphorylation, which enhances kinase activity and leads to UPR-targeted gene transcription. This process is essential for proper protein folding and ER function restoration. Additionally, IRE1α is involved in Jun N-Terminal Kinase (JNK) phosphorylation, thereby activating the cellular MAP kinase pathway, which further underscores IRE1α′s importance in cellular signaling and stress response mechanisms.

Knocking-down Ire1, I validated this antibody for WB (strong band, disappears with siRNA) but not for IF (formaldehyde-fixed, strong signal, stays the same with siRNA). How did you validate this antibody in IF? Do you recommend an antigen retrieval step?

We have tested this antibody in immunofluorescence cell stainings with the standard protocol you can find on our website: https://www.scbt.com/resources/protocols/immunofluorescence-cell-staining If you are performing fixation with formaldehyde, antigen retrieval may be necessary. Can this antibody detect both phosphorylated and non-phosphorylated forms of IRE1-α?

Thank you for your question. IRE1α Antibody (B-12): sc-390960 was aised against amino acids 371-560 of IRE1α of human origin. It recognizes both the phosphorylated and the non-phosphorylated IRE1α.

Does this antibody recognize Ire1 from Drosophila?

Thank you for your question. IRE1α Antibody (B-12): sc-390960 is recommended for detection of IRE1α of mouse, rat and human origin by WB, IP, IF and ELISA. There is currently no data on the antibody’s reactivity in Drosophila. We have not received any customer feedback indicating reactivity in other species than mouse, rat, or human. Please contact our technical service for further information or to arrange for a free sample of the antibody to test whether it would be suitable for your application.

Rated 5 out of 5 by from Really nice band!We checked the antibody in MDCK cells (canine). We used the antibody in 1:1.000 dilution in bovine serum albumin (1% in TBST), and the secondary antibody in 1:10.0000 dilution. We got a great band, with the expected molecular weight.
