    CAPON Antibody (C-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects CAPON protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-CAPON antibody (C-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CAPON, or carboxy-terminal PDZ ligand of nNOS, plays a crucial role in the regulation of neuronal nitric oxide synthase (nNOS) by selectively binding to the 100 amino acid PDZ domain of nNOS, while not interacting with endothelial or inducible NOS. This interaction is vital as CAPON sequesters nNOS in the cytosol, thereby influencing the biosynthesis of nitric oxide (NO), a key neurotransmitter involved in various physiological processes. The proper functioning of nNOS depends on association with synaptic proteins such as syntrophin, postsynaptic density protein 95 (PSD95), and PSD93, which facilitate transport to the plasma membrane where nNOS becomes catalytically active through NMDA-receptor mediated calcium channels. CAPON regulates nNOS association with PSD95 and PSD93 by competing for binding to the PDZ domain, thus preventing translocation and activation of nNOS. This regulatory mechanism maintains the balance of nitric oxide signaling in the nervous system, highlighting CAPON′s importance in synaptic function and neurophysiology.

    Rated 5 out of 5 by from The antibody has high specificityI used cell lysate from HT22 to do western blotting, and loaded about 20μg protein to every lane. The dilution of CAPON (C-9) was 1:200. The band was clear and had no other bands. The antibody is good.
