SEE ALSO...

α1A-AR Antibody (4D8) is a mouse monoclonal IgG1 kappa light chain antibody that detects α1A-AR protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-α1A-AR antibody (4D8) is available as a non-conjugated format. α1A-AR plays a crucial role in the sympathetic nervous system by mediating catecholamine effects such as epinephrine and norepinephrine, which regulate cardiovascular functions. α1A-AR couples with Gq/11 proteins, leading to vasoconstriction and increased blood pressure, influencing vascular tone and cardiac output. α1A-AR expression spans across heart, brain, liver, and prostate tissues, where alternative splicing of ADRA1A gene results in multiple isoforms with unique C-termini showing distinct activation patterns. α1A-AR transcripts in heart, liver, and prostate, ranging from 2.0 to 6.0 kb, highlight significant roles in physiological processes and potential implications in pathophysiological conditions.

How selective is this antibody for alpha-1A? We need to address the selectivity issue in light of Michel et al. (2009) "How reliable are G-protein-coupled receptor antibodies?" NAUNYN-SCHMIEDEBERGS ARCHIVES OF PHARMACOLOGY 379: 386-388.

Thank you for your question. The manuscript you mention recommends validating by IHC, but I would like to aslso suggest that also using siRNAs and CRISPR followed by a WB would also provide validation results to determine the reliability of the antibody. You can find recommended gene silencing products on the antibody product block: https://www.scbt.com/scbt/product/alpha1a-ar-antibody-4d8?requestFrom=search If you have any further questions, please contact Technical Service by phone, (800)-457-3801 option 2, email <scbt@scbt.com>, or by live chat directly on our website, www.scbt.com.

For Western Blot, is it recommended to use denatured or non-denatured conditions with α1A-AR (4D8): sc-100291 antibody?

Asked by: Germaine
Thank you for your question. We recommend this antibody for use in denatured Western Blot conditions. It has not been validated for use in non-denatured conditions. Please contact our Technical Service Department for further details or inquiries.
