SEE ALSO...

IFN-γ Antibody (E-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects interferon gamma (IFN-γ) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). IFN-γ (E-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IFN-γ plays a crucial role in the immune response, acting as an antiviral and antiparasitic agent produced primarily by CD4+/CD8+ lymphocytes and natural killer cells upon activation by antigens, mitogens, or alloantigens. IFN-γ modulates T cell growth and differentiation while inhibiting B cell proliferation, thereby influencing the overall immune response. Various cytokines can induce IFN-γ synthesis, including interleukin-2, fibroblast growth factor, and epidermal growth factor, highlighting IFN-γ′s importance in immune regulation. Structurally, active IFN-γ exists as a homodimer, with each subunit comprising six helices, and this dimeric configuration is stabilized by non-covalent interactions at the helices′ interface. IFN-γ precursor consists of 166 amino acids, which includes a 23 amino acid secretory sequence, and functional diversity is further enhanced by variable glycosylation and the formation of dimers and tetramers under non-denaturing conditions.

What application is the blocking peptide sc-373727 P appropriate for?

Asked by: Trav11
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

Rated 1 out of 5 by from Holy (and expensive) waterTried the free IFNgamma (E-10) in Western blot (1:100 as suggested by datasheet) with olfactory mesenchymal stem cells (oMSC) and mouse cortex protein extracts to check for signal in these two models. Not a single band was detected. Everything was done properly. I have tested 8 (eight!) mouse monoclonal antibodies from Santa Cruz and not a single one gave any band except one that gave an aspecific signal (I would like to upload a picture, but it is not allowing me). I will not loose more time with them and I strongly advice you to do the same. Good luck, fellow researchers.
