ephrin-B2 Antibody (F-2) is a mouse monoclonal IgG2a kappa light chain antibody that detects ephrin-B2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ephrin-B2 Antibody (F-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ephrins, which serve as ligands for Eph receptors, are critical cell-surface proteins categorized into ephrin-A and ephrin-B based on structural and functional characteristics. ephrin-B2 is particularly significant due to a role in vascular development; ephrin-B2 is a transmembrane protein containing conserved cytoplasmic tyrosine residues that undergo phosphorylation upon binding to EphB receptors. This phosphorylation initiates bidirectional signaling pathways influencing cell positioning and tissue organization during embryogenesis. ephrin-B2 is expressed on arterial endothelial cells, while EphB4 receptor is found on venous endothelial cells, facilitating communication necessary for proper vascular morphogenesis. ephrin-B2 and EphB4 interaction is essential not only for normal embryonic vascular development but also plays a pivotal role in tumor angiogenesis, making ephrin-B2 a significant target in cancer therapeutics.

Can ephrin-B2 (F-2): sc-398735 monoclonal antibody be used to detect the extracellular fragments of EphrinB2

Thank you for your question. The epitope for ephrin-B2 (F-2), sc-398735, maps within amino acids 168-235, however the exact epitope has not been mapped. The extracellular region for this protein is amino acids 28-229, so chances are very good the epitope is within the extracellular region.

I have got sc-398735. However, there is no result in IF. Is the second antibody (such as sc-398735 AF488 and sc-398735 AF647 ) must be used?

Thank you for your question. It is not necessary to use the conjugated version of this mouse monoclonal primary antibody, ephrin-B2 (F-2): sc-398735, for immunofluorescence (IF). You can use a conventional anti-mouse IgG secondary antibody for detection, or one of our mouse IgG kappa light chain binding proteins (e.g., m-IgGκ BP-CFL 488: sc-516176). I have 2 questions: 1. Is it antibody can be used for IHC(P)? we have "Dako EnVision Dual Link System-HRP" as the second antibody. 2. If it can be used in IHC(P), which isotype control antibody is suggested? Thank u so much!

Thank you for your question. We have not yet tested this mouse monoclonal, ephrin-B2 Antibody (F-2): sc-398735, on formalin-fixed paraffin-embedded tissue sections, or for IHC(P) so we do not know if the antibody is suitable for this application. If you want to test the antibody for IHC(P), the antibody has an isotype of mouse monoclonal IgG2a so the corresponding isotype control is our, normal mouse IgG2a: sc-3878. https://www.scbt.com/scbt/product/normal-mouse-igg2a

Can ephrin-B2 (F-2): sc-398735 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, ephrin-B2 (F-2): sc-398735 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 1 out of 5 by from not good for immune-staining in brain sectionI used this trial recently and do IF in mice brain section but not good with the solution 1:100 and 1:50. Very little signals were seen. I use this as a marker for brain arterial vessel, it should be a lot under the scope.
