PP1α Antibody (G-4) is a mouse monoclonal IgG2b kappa light chain antibody designed to detect the full-length human phosphatase PP1α protein, also known as PPP1CA or Protein Phosphatase 1α. PP1α monoclonal antibody (G-4) is reactive across multiple species, including mouse, rat, human, canine, bovine, and porcine, making it versatile for various applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Available in both non-conjugated form and with conjugations like agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants, anti-PP1α antibody (G-4) offers flexibility for diverse experimental needs. Phosphatase PP1α is a critical serine/threonine-protein phosphatase localized predominantly in both the nucleus and cytoplasm of eukaryotic cells, where PP1α plays an essential role in regulating various cellular functions including cell division, homeostasis, and apoptosis through the dephosphorylation of target proteins. The precise localization of PP1α within the cell is vital for function, as PP1α ensures the correct modulation of phosphorylation states in specific cellular compartments, thereby maintaining cellular balance and responding to signaling cues. PP1α operates as part of the PP1 holoenzyme complex, interacting with regulatory and variable subunits to achieve substrate specificity and regulation. PP1α activity is tightly controlled by inhibitory proteins such as NIPP-1, and regulation through phosphorylation by kinases like cAMP-PK and casein kinase II is crucial for activating or inhibiting phosphatase activity as needed. Understanding and detecting PP1α localization and activity with PP1α (G-4) antibody is therefore fundamental for studies investigating cellular signaling pathways, protein interactions, and the mechanisms underlying various physiological and pathological processes.

Was this product validated for isotope specificity, i.e. does it detect only PP1alpha and not PP1beta and PP1gamma?

Asked by: cstech
PP1α Antibody (G-4) is specific for an epitope mapping between amino acids 297-326 at the C-terminus of PP1α of human origin. It does not detect PP1β or PP1γ. For an overview of all antibodies that detect PP1 or one of its isotypes, just follow this link: https://www.scbt.com/scbt/browse/pp1-Antibodies/_/N-nb1lg9 Please contact technical service if you have further questions.

What application is the blocking peptide sc-271762 P appropriate for?

Asked by: Cweed
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
