PKC α Antibody (H-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects PKC alpha protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). PKC α Antibody (H-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. PKC alpha protein, a member of the protein kinase C (PKC) family, plays a crucial role in regulating numerous cellular functions such as cell growth, differentiation, gene expression, and hormone secretion, making PKC alpha vital for maintaining cellular homeostasis. PKC alpha is primarily located in the cytoplasm and translocates to the cell membrane upon activation, which is essential for PKC alpha′s role in signal transduction pathways. This translocation is triggered by diacylglycerols (DAG) and tumor-promoting phorbol esters, which bind to and activate PKC alpha, allowing PKC alpha to phosphorylate target proteins and modulate various cellular responses. The diverse expression patterns of PKC isoforms across different tissues highlight PKC alpha′s importance in specific signaling pathways, and PKC alpha′s unique cofactor dependencies further underscore PKC alpha′s distinct regulatory mechanisms within the PKC family.

does this product work on zebrafish embryo?any suggestion for zebrafish use,the product SC-216 is not available now!

Thank you for your question. We are not aware of any publications citing use of this mouse monoclonal antibody, PKC α (H-7): sc-8393, in zebrafish samples. The antibody is raised against a peptide that does share significant (76%) sequence identity with PKC α in zebrafish, so there is a good chance that it would work in this species. For a limited time, we are offering a free 10 µg sample of this antibody. Please contact our Technical Service Department if you would like to request a sample to test in zebrafish.

Rated 5 out of 5 by from Dendritic morphology of bipolar cells in wild type10 to 15 weeks old mice were anesthetized with isoflurane (Vetflurane®k, Virbac) and killed by cervical dislocation. Vertical sections: The following steps were conducted at room temperature if not stated otherwise. Eyes were quickly removed from the eye socket, opened at the scleral-corneal rim and immersed for 10 min with 4% paraformaldehyde (PFA) in 1X phosphate-buffered saline (1X PBS, pH 7.4). Cornea, lens and vitreous were removed. Eye cups were then fixed with 4%PFA/1X PBS for 20 min, washed four times with 1X PBS and cryoprotected by a graded sucrose series: 10% sucrose in 1X PBS for 1 h, 20% sucrose in 1X PBS for 1 h and 30% sucrose in 1X PBS overnight at 4 °C. Eyecups were orientated along the dorsoven-tral axis, embedded in OCT Medium (Tissue-Tek O.C.T Compound; Sakura Finetek, Tokyo, Japan) and frozen in liquid nitrogen. Vertical sections (16 μm) were cut on a cryostat (Leica Microsystems, Wetzlar, Germany), mounted on gelatine coated slides and stored at − 20 °C. For immunofluorescence experiments, sections were washed three times in 1X PBS-T (1X PBS + 0.1% Triton X-100, Sigma-Aldrich, St. Louis, MO, USA), blocked for 1 h in 1X PBS-T containing 1% bovine serum albumin (BSA, Sigma-Aldrich, A7030) and incubated overnight at 4 °C with primary antibodies diluted in 1X PBS-T at concentrations listed in Supplementary table 1. After wash-ing three times with 1X PBS-T, sections were incubated with the secondary antibodies (Supplementary table 2) for one. Additional washes preceded the counterstaining with DAPI (1:10,000; Sigma, D-9542) and eventually the sections were mounted using Poly/Mount (Polysciences, Inc., Warrington, PA, USA)

We found PKA-A canbe used assit for lung cancer diagnosis.We recommend the use of 1:300 for immunohistochemistry.
