Bcr Antibody (A-1) is a mouse monoclonal IgG1 kappa light chain antibody, specifically engineered to recognize an epitope mapping between amino acids 2-29 at the N-terminus of the human Bcr (Breakpoint Cluster Region) protein, also known as Renal Carcinoma Antigen NY-REN-26. The Bcr protein plays a crucial role in cellular signaling by functioning as a GTPase-activating protein (GAP) for Rac1 p21, which is essential for regulating cytoskeletal dynamics, cell migration, and proliferation, thereby maintaining normal cellular functions and preventing uncontrolled cell growth. Located predominantly in the cytoplasm, the precise cellular localization of Bcr is vital for its interaction with other signaling molecules and its role in maintaining cellular architecture. Structurally, Bcr contains multiple domains, including the GAP domain, which is critical for its interaction with Rac1 and modulation of its activity, ensuring proper regulation of downstream signaling pathways. Post-translational modifications, such as phosphorylation, significantly influence Bcr′s activity and interactions, thereby affecting processes like cell adhesion and movement. Bcr interacts with several key proteins, including c-Abl, forming the Bcr/Abl fusion protein in the Philadelphia chromosome translocation commonly found in chronic myelogenous leukemia (CML), as well as adaptor proteins like Crk, which are essential for propagating downstream signaling events. The Bcr gene is located on chromosome 22 and its fusion with the c-Abl proto-oncogene on chromosome 9 results in the constitutive tyrosine kinase activity of the Bcr/Abl fusion protein, driving the proliferation of leukemic cells. Anti-Bcr antibody (A-1) effectively detects the Bcr protein in mouse, rat, and human samples, as well as in additional species including canine, making it versatile for various research applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA).
