Sox-9 Antibody (E-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects Sox-9 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Sox-9 (E-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Sox-9 plays a crucial role in the regulation of chondrogenesis and is essential for cartilage development and male gonad formation. Is this product appropriate for flow cytometry? Have you got citations of people successfully using it?

Thank you for your question. Fluorescent conjugates of this antibody can be used for flow cytometry. As of yet, we have not received any published data on flow cytometry use.

Can the conjugated version of this antibody (546) be used in combination with other secondary antibodies for double-staining? I haven't used many mouse-on-mouse stains and I know that can be problematic but mouse is listed under reactive species.

Thank you for your question. We do not recommend using a secondary antibody or binding protein with a primary antibody that is already conjugated. Steric hindrance may negatively influence your results. Most samples and tissues are unproblematic when using a mouse primary antibody and anti-mouse secondary detection. When working with tissues make sure to remove all blood from the sample to prevent the secondary from picking up endogenous IgG. Using a direct conjugate (without a secondary) will circumvent this issue entirely.

Has anyone tried with IHC before? I have got mice testes samples which are paraffin fixed. I tried doing it but there isn't any signal.

Thank you for your question. We have not yet tested this antibody in formalin-fixed paraffin-embedded tissue sections, and so IHC(P) is not a recommended application at this time. We have successfully used the antibody on formalin-fixed cells for IF. In addition, there are several publications citing use of this antibody for IHC staining. For example, PMID's: 25917841 and 30244259.

Is this product acceptable for ChIP/ChIP-seq applications?

I used this antibody for IF in rat cartilage cryosections but no staining was observed. Could you please recommend a protocol for this? Which is the best fixative and permeabilization agent? Is antigen retrieval necessary? Thanks.

Thank you for your question. I have provided a link to our recommended protocol for immunofluorescent staining of frozen tissue sections. We recommend fixing slides in cold acetone for 10 minutes. A permeabilization agent should not be necessary if fixing in acetone. If you are using a formalin-based fixative, then permeabilization with Triton X-100 may be needed. I would also recommend reviewing the methods in the following publication on PubMed (PMID: 25917841) which cites use of this Sox-9 Antibody (E-9): sc-166505 for IHC in rat tissue. https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

What is the recommended fixation method for IF with this Sox-9 (E-9): sc-166505?
