FKHRL1 Antibody (D-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects FKHRL1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). FKHRL1 (D-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FKHRL1 protein, also known as FOXO3 or FOXO3A, is a crucial transcriptional activator composed of 673 amino acids that plays a significant role in regulating cellular processes such as apoptosis and cell cycle progression. FOXO3 regulation occurs through post-translational modifications, particularly phosphorylation by serine/threonine kinase Akt1. When phosphorylated at Threonine 32 and Serine 253, FOXO3 is sequestered in the cytoplasm through association with 14-3-3 proteins, inhibiting transcriptional activity. Under growth factor withdrawal conditions, FOXO3 undergoes dephosphorylation, translocating to the nucleus where FOXO3 activates pro-apoptotic gene expression. This dynamic regulation of FOXO3 is vital for maintaining cellular homeostasis and responding to stress signals, making FOXO3 an important target for research in cancer and metabolic diseases.
