SEE ALSO...

Aprataxin Antibody (B-12) is a mouse monoclonal IgG2a kappa light chain antibody that detects Aprataxin of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Aprataxin antibody (B-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Aprataxin is primarily located in the nucleus, specifically within the nucleoplasm and nucleolus, where Aprataxin plays a crucial role in DNA single-strand break repair. This nuclear localization enables Aprataxin to effectively mediate protein-protein interactions with other molecules involved in cellular response to DNA damage, thereby maintaining genomic stability. Aprataxin belongs to the histidine triad (HIT) superfamily and features three conserved domains: an N-terminal forkhead-associated (FHA) domain that facilitates these interactions, a HIT domain similar to Hint, and a C-terminal zinc finger domain. Mutations in the APTX gene, which encodes for Aprataxin, can lead to protein destabilization and are associated with a rare neurological disorder known as ataxia-oculomotor apraxia, characterized by abnormal head and eye movements. These mutations often affect the HIT domain or result in truncation before the zinc finger domain, underscoring Aprataxin′s importance in cellular repair mechanisms and neurological health.

Can Aprataxin (B-12): sc-374108 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: cjMara
Thank you for your question. Yes, Aprataxin (B-12): sc-374108 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
